Somatropin
Indications/Uses
Long-term treatment of childn w/ growth disturbance caused by insufficient secretion of growth hormone. Turner syndrome, chronic renal insufficiency, born small for gestational age, Prader-Willi syndrome, pre pubertas idiopathic short stature. Improvement of body composition in childn w/ Prader-Willi syndrome. Replacement therapy in adults w/ growth hormone deficiency.
Dosage/Direction for Use
Adult Starting Dose: 0.15-0.3 mg (0.45-0.9 IU) daily. Final dose should be individually titrated as needed w/ respect to age & gender. Maintenance dose seldom exceeds 1.3 mg (4 IU) daily. Women may require higher doses than men. Childn Growth hormone deficiency 0.025-0.035 mg/kg daily. Turner syndrome & chronic renal insufficiency 0.045-0.05 mg/kg daily. Prader-Willi syndrome 0.035 mg/kg daily. Small for gestational age 0.035-0.067 mg/kg daily. Pre-pubertas idiopathic short stature Up to 0.067 mg/kg daily.
Contraindications
Hypersensitivity. Evidence of activity of a tumour, or of growth tumour. Closed epiphysis. Acute catabolism including pre- & post-op treatment, critically ill & burn patients.
Special Precautions
Severe obesity, history of resp impairment or sleep apnea or unidentified resp infection. Myositis. Relapse of malignancy of a malignant disease, DM, hypothyroidism during GH treatment, endocrine disorders, including GH deficiency. Excessive glucocorticoid therapy will inhibit the growth promoting effect of human growth hormone. Patients w/ coexisting ACTH deficiency should have their glucocorticoid replacement dose carefully adjusted to avoid an inhibitory effect on growth. Pregnancy & lactation.
Adverse Reactions
Adult: Oedema peripheral, musculoskeletal stiffness, arthralgia, myalgia & paraesthesia. Inj site reactions eg, itching, lumps, redness & lipoatrophy.
Drug Interactions
May increase the clearance of compd metabolized by cytochrome P4503A4 (eg, sex steroids, corticosteroids, anticonvulsants & cyclosporin).
